Flammulina velutipeshas been reported to be beneficial to learning and memory capability, but the mechanisms underlying this remain unclear. In the present study, Morris water maze test and biochemical analysis for hippocampus and cerebral cortex of rats were performed to evaluate the effects of polysaccharides from F. velutipes (FVP) on scopolamine-induced learning and memory impairment. Results suggested that FVP significantly decreased the escape latency and total swimming distance of rats in hidden platform test, and increased the numbers of crossing platform and distance of swimming around platform of rats in probe test. Biochemical examination revealed that FVP significantlyelevated SOD and GSH-Px activities, and acetylcholine (ACh), dopamine (DA), 5-hydroxytryptamine (5-HT), and norepinephrine (NE) levels, especially the increased ACh content in hippocampus owed to the increased acetylcholine acetyltransferase activity and decreased acetylcholinesterase activity. Moreover, learning and memory associated signaling pathways were activated by FVPelevating the expression of Connexin 36 and p-CaMK II. The present results conclusively proved that FVP ameliorated learning and memory impairments by mainly regulating the expression of CaMK II, activating the activities of ChAT and AChE and then normalizing the level of Ach.
